正小麦蓝矮病(Wheat blue dwarf,WBD)是我国西北麦区一种重要植原体病害,在我国西部地区危害严重。该病害由异沙叶蝉(Psammotettix alienus L.)专化性传播,介体传毒成为病害流行的重要中心环节[1]。本实验室前期通过免疫荧光标记研究发现WBD植原体免疫膜蛋白(immunodominant membrane protein, IMP)与介体异沙叶蝉肌动蛋白互作,说明IMP在植原体传播和致病过程中起关键作用。  相似文献

采用过氧化氢刺激lager型啤酒酵母,Tadpoling法筛选细胞活力恢复较快突变菌,根据各菌株线粒体膜电位、胞内氧自由基(ROS)水平筛选获得4株活性较高菌株。比较突变菌与原始菌传代发酵性能、胞内ROS、活力指标及每一代细胞死亡率水平,最终获得一株抗氧化能力提高且活力稳定性较高啤酒酵母菌株。分析验证突变菌线粒体DNA修复相关基因MHR1发现,该基因发生部分碱基突变。  相似文献

Fish were fed a single‐strain yeast fraction (SsYF; 2 g/kg) or a multistrain yeast fraction (MsYF; 0.8 g/kg) for 10 weeks. The results demonstrated significant (p ≤ 0.03) elevations in weight gain, specific growth rate, protein efficiency ratio, and feed conversion ratio in fish fed the yeast fraction‐supplemented diets. In the distal intestine, a significant elevation in microvilli density was observed after 5 and 10 weeks of dietary supplementation with MsYF and SsYF, respectively, compared to control fed fish (p < 0.001). A significant elevation (p = 0.02) in the perimeter ratio was observed in fish fed diets supplemented with the yeast fractions. After 10 weeks of feeding on the experimental diets, Rt‐qPCR demonstrated a significant downregulation (p < 0.05) in the stress response genes, heat‐shock protein 70 (hsp70) and proliferating cell nuclear antigen (pcna), in fish fed diets supplemented with the yeast fractions. Significant (p < 0.05) elevations in interleukin 1‐beta (il1β) and interleukin‐10 (il10) gene expression were observed in fish fed diets supplemented with the MsYF compared to the other dietary groups. These findings suggest that feeding an MsYF specifically at a lower incorporation rate < 1 g/kg, compared to a commercial SsYF at 2 g/kg, is effective in improving the intestinal health status and growth performance of European seabass.  相似文献

从蜂蜜、葡萄、面包3种原料中提取天然酵母进行梯度驯化,再通过紫外诱变育种,筛选出具有耐高糖性质的酵母并对其相关特性进行研究,将筛选出的菌株应用于黄酒酿造过程。结果表明,筛选出的酵母可耐受的糖度、pH值、酒精度分别为40%,1.5和10%Vol,将其应用于黄酒发酵中,经过7 d发酵后,菌株降糖能力较传统菌株提高了20 g/L,酒精度提高了0.4%Vol,糖类物质转化率达到11%。采用自然驯化和紫外诱变育种相结合,得到适用于黄酒酿造生产的耐高糖酵母菌株。  相似文献

A 60‐day feeding trial was carried out to evaluate the effects of chromium yeast (CY), tributyrin (TB) and bile acid (BA) on the snakehead (Channa argus). The control (CT) group was fed the basal diet, and the experimental groups were fed 200 mg/kg CY (concentration: 1,000 mg/kg), 2.5 g/kg TB (purity: 45%) or 1 g/kg BA (purity: 30%) in addition to the basal diet. The addition of CY, TB and BA statistically increased the weight gain rate, and specific growth rate, whereas the feeding rate and feed conversion ratio were significantly reduced (p < 0.05). When compared with the CT group, TB elevated the serum glutathion peroxidase (GSH‐Px) activity; and BA reduced low density lipoprotein cholesterol (LDL‐C) contents significantly (p < 0.05). The three feed additives reduced intestinal malonaldehyde (MDA) levels dramatically. Tributyrin significantly enhanced intestinal trypsin and pepsin activities (p < 0.05). The activity of fructose‐1,6‐bisphosphatase and pyruvate carboxylase in the group fed CY was significantly lower than in the CT group. In conclusion, dietary inclusion of the three additives enhanced growth performance of C. argus. Dietary supplementation of CY affected protein, glucose and lipid metabolism. Diets with TB levels improved intestinal digestive enzyme activity and antioxidant capacity. The addition of BA improved glycolipid metabolism.  相似文献

The purpose of this study was to evaluate the effect of yeast with bacteriocin from Ruminococcus albus 7 (albusin B) on physiological state and production performance of laying hens. One hundred and twenty 26‐week‐old Single Comb White Leghorn (Hyline) laying hens were assigned into five groups including: (i) control group, (ii) yeast control (YC), (iii) 0.125% yeast with bacteriocin (0.125B), (iv) 0.25% yeast with bacteriocin (0.25B) and (v) 0.5% yeast with bacteriocin (0.5B). All supplements were added to the experimental diets of the hens from 26 to 46 weeks of age. Samples were collected every 4 weeks. Blood samples were collected from the wing vein for blood biochemical parameters assay, and faecal samples were collected by swab for the microbiota test. The egg production performance was recorded daily, and fresh eggs were collected for quality test. The blood biochemical assay results indicated that the addition of yeast with bacteriocin decreased the AST (aspartate aminotransferase) activity and it also affects the lactate concentration in laying hen blood. The result of egg quality indicated that yeast with bacteriocin supplementation had no effect on the mass of yolk and the strength of eggshell, but it had positive effect on the laying performance under hot environment. Low concentration bacteriocin (0.125B) supplementation could decrease total yolk cholesterol. The faecal microbiota result indicated that the supplementation of bacteriocin increased the lactobacilli counts. The yeast with bacteriocin supplementation significantly decreased the clostridia counts under hot environment condition, especially in hens receiving 0.25B. Combining the data from clinic chemistry, faecal microbiota, egg production and egg quality, the 0.25B supplementation may result in the best physiological parameter and egg production performance of laying hen.  相似文献

枝干病害葡萄溃疡病近年来严重限制了葡萄产业发展。研究表明葡萄蔗糖转运蛋白参与寄主植物和病原菌的互作过程。为解析蔗糖转运蛋白VvSUC12在葡萄免疫反应过程中的功能,本研究克隆了VvSUC12基因翻译起始位点上游1 500 bp的启动子区,通过生物信息学分析发现该区域包含4个Dof转录因子结合序列(A/TAAAG)及多种激素调节与防御相关的顺式元件。实时荧光定量分析显示,接种可可毛色二孢菌显著诱导VvSUC12和VvDof19基因的表达。通过酵母单杂交实验筛选得到与VvSUC12启动子互作的转录因子VvDof19。酵母双杂交实验证实VvDof19具有自激活活性;进一步通过烟草瞬时表达发现Dof19-GFP的相对GUS活性约为对照GFP的9倍,表明转录因子VvDof19能够激活VvSUC12基因的表达。研究结果为深入研究蔗糖转运蛋白在葡萄免疫反应中的功能奠定基础。  相似文献

A 6‐week broiler study was conducted to evaluate whether subjecting the intestinal microflora of broilers to the effect of weekly variations in feed ingredients could be ameliorated by the inclusion of yeast‐derived feed additives: a yeast cell wall extract (YCW), live yeast culture (LY) or their combination (YCW + LY). Recent changes in ingredient prices have motivated producers to formulate diets not necessarily based primarily on corn and soya bean meal. Intestinal microflora in birds can vary significantly based on the ingredient composition of their diet, and the make‐up of the flora can influence overall bird performance. Within the three nutrient phases of this study, birds were fed either a traditional corn–soya ingredient profile or a variable‐ingredient regimen, which had weekly changes in the ingredient composition. There were consistent ameliorative effects of the yeast treatments in both the corn–soya and the variable‐ingredient groups throughout all 6 weeks, with the YCW + LY combination showing a reduced effect when compared to either product fed alone. The effectiveness of YCW and LY on ameliorating the effects of weekly ingredient variations appeared most effective during the starter and grower phases, but was less significant during the sixth week.  相似文献
